چکیده

SA4503 [1-(3,4-dimethoxyphenethyl)-4-(3-phenylpropyl)piperazine dihydrochloride] has been described as a new, selective sigma ( ) receptor agonist with a potential antidepressant activity [8, 9, 17]. It has been reported that SA4503 shows an antidepressant-like activity in mice [8, 17]. Similar effects were observed after the administration of (+)-pentazocine (another receptor agonist) and DTG (a / agonist) to mice and rats. Moreover, those antidepressant-like effects were antagonized by the receptor antagonist, NE-100 [8, 9, 16, 17]. Our previous studies indicated that SA4503 decreased the immobility time in the forced swimming test in rats and showed a synergistic effect with imipramine in that model. When given repeatedly (14 or 21 days), SA4503 induced effects similar to those produced by many clinically active antidepressant drugs (ADs), e.g. it enhanced the D-amphetamineor quinpirole-induced hyperactivities in rats, and did not change the D-amphetamine stereotypy (in preparation). It was demonstrated that ADs administered repeatedly increased the responsiveness of the -adrenergic system (sensitivity of postsynaptic -adrenergic receptors) at both behavioral and biochemical levels (e.g. they increased the clonidine-induced Polish Journal of Pharmacology ! High occupancy of sigma1 receptors in the human brain after single oral administration of donepezil: a positron emission tomography study using [11C]SA4503.

The acetylcholinesterase (AChE) inhibitor donepezil is also a sigma1 receptor agonist. We examined whether donepezil binds to sigma1 receptors in the living human brain after a single oral administration. Dynamic positron emission tomography (PET) data acquisition using the selective sigma1 receptor ligand [11C]SA4503 was performed to evaluate quantitatively the binding of [11C]SA4503 to sigma1...

The synergistic effect of selective sigma receptor agonists and uncompetitive NMDA receptor antagonists in the forced swim test in rats.

Data acquired to date show that some sigma receptor ligands reveal "antidepressant-like" activity in the forced swim test in mice and rats. Moreover, our preliminary results suggested that joint administration of sigma receptor ligands and amantadine (AMA, a glutamatergic/NMDA receptor antagonist) caused a positive interaction in the Porsolt test in rats, as had already been observed in the cas...

Age-related changes of the binding of [3h]SA4503 to sigma1 receptors in the rat brain.

We have recently developed 1-([3-O-methyl-11C]3,4-dimethoxyphenethyl)-4-(3-phenylpropyl) piperazine ([11C]SA4503) as a selective radioligand for mapping sigma1 receptors in the brain by positron emission tomography (PET). In the present short communication we evaluated the age-related changes of the binding of this ligand to sigma1 receptors in Fisher-344 rats (1.5-, 6-, 12-, and 24-month-old) ...

[Cardioprotective effect of the selective sigma-1 receptor agonist, SA4503].

We previously reported that the sigma-1 receptor is down-regulated in cardiomyocytes following heart failure in transverse aortic constriction (TAC) mice. In this review, we summarized the anti-hypertrophic action of selective sigma-1 receptor agonist, SA4503 in the hypertrophied cultured cardiomyocytes and discussed its possible mechanism of cardioprotection. Treatment with SA4503 (0.1-1 μM) d...

Possible Involvement of Glutamatergic, Adrenergic and Dopaminergic System in Methylphenidate - induced Motor Activity and Mood-related Alterations in Rats

Background and Objective: Methylphenidate (MPH), as a central nervous system stimulant, is often used to manage hyperactive disorders. The literature is scarce regarding the behavioral consequences of chronic MPH treatment and the role of involved receptors. Thus, in the current study involved receptors in MPH induced-anxiety, depression and motor activity disorders were evaluated. Materials a...
